conclusions nucleic acid amplification tests (naats) are very appropriate in detection of infected individuals. detection techniques such as naats are independent of bacterial viability, and have a potential to limit false negative samples, therefore, in our country, the application of different laboratory diagnosis methods including naats with culture as gold standard for determination antimicrobial susceptibility is essential. results among 1100 endocervical swabs examined, 13 (1.18%) samples had positive results by polymerase chain reaction (pcr) on neisseria gonorrhea ccpb gene. all endocervical swabs culture had negative results for neisseria gonorrhea. 84 (7%) of the women had vaginal discharge, in whom pcr on endocervical swabs of these individuals had negative findings. patients and methods endocervical swabs specimens for this study were obtained from 1100 pregnant women who presented to shiraz (iran) hospitals from 2009 to 2011. in the present investigation we used conventional agar-based culture method, and nucleic acid amplification test (naat) of ccpb gene for detection of neisseria gonorrhea in endocervical swabs samples collected from pregnant women studied. from each pregnant woman two endocervical swabs were taken: one swab placed in tubes containing phosphate buffered saline for polymerase chain reaction, and the other to inoculate on culture media. objectives in the present investigation, we used conventional agar-based culture method, and nucleic acid amplification of ccpb gene for detection of neisseria gonorrhea in endocervical swabs samples collected from pregnant women studied background neisseria gonorrhea is the etiological agent of the sexually transmitted disease (std) gonorrhea, and primarily infects the mucous membranes of the urethra, endocervix, pharynx or rectum of females which may result in substantial morbidity. n. gonorrhea also causes disseminated infection, with complications that may result in ectopic pregnancy, tubal infertility, chronic pelvic pain or maternal transmission of gonorrhea, and also increases susceptibility to hiv.

Sensitivity and Specificity of Nucleic Acid Sequence-Based Amplification Method for Diagnosis of Cutaneous Leishmaniasis

Abstract Background and Objective: Culture, microscopic method is a gold standard method for identification of Lishmania parasite. The use of Molecular methods such as RT- PCR compared to microscopic methods has a higher sensitivity and specificity however, it is not widely used due to its expensive equipment and the time requested. The use of nucleic acid sequence based amplification (NASBA) ...

COMPARISON OF CULTURE WITH POLYMERASE CHAIN REACTION FOR DETECTION OF UREAPLASMA UREALYTICUM IN ENDOCERVICAL SPECIMENS

Background: Ureaplasma urealyticum is known as a sexually transmitted agent, causing mainly urethritis, pelvic int1ammatory disease, spontaneous abortion, pyelonephritis, infertility, stillbirth, low birth weight neonatal meningititis, and neonatal pneumonia. U. urealyticum infections not only jeopardize fertility but also pose a risk for infertility treatment and resulting pregnancies. Dia...

Molecular Assay for Detection of Ciprofloxacin Resistance in Neisseria gonorrhoeae Isolates from Cultures and Clinical Nucleic Acid Amplification Test Specimens.

We developed a real-time PCR assay to detect single nucleotide polymorphisms associated with ciprofloxacin resistance in specimens submitted for nucleic acid amplification testing (NAAT). All three single nucleotide polymorphism (SNP) targets produced high sensitivity and specificity values. The presence of ≥2 SNPs was sufficient to predict ciprofloxacin resistance in an organism.

Evaluation of Nucleic Acid Sequence Based Amplification (NASBA) and Reverse Transcription Polymerase Chain Reaction for Detection of Coxsackievirus B3 in Cell Culture and Animal Tissue Samples

Enteroviruses are the causative agents of a number of diseases in humans. Group B coxsackieviruses are believed to be the most common viral agents responsible for human heart disease. Genomic data of enteroviruses has allowed developing new molecular approaches such as Nucleic Acid Sequence Based Amplification (NASBA) for detection of such viruses. In this study, coxsackievirus B3 (CVB3) was de...

Comparing first-void urine specimens, self-collected vaginal swabs, and endocervical specimens to detect Chlamydia trachomatis and Neisseria gonorrhoeae by a nucleic acid amplification test.

We set out to determine the prevalences of Chlamydia trachomatis and Neisseria gonorrhoeae by ligase chain reaction as well as to determine the prevalence of Trichomonas vaginalis by culture in a large and diverse national sample of non-health-care-seeking young women entering the military; we also sought to compare the abilities of three different techniques of collecting specimens (first-void...
